Despite their significance, until now, no book has been dedicated to hydrolysis and hydrolases in the metabolism of drugs and other xenobiotics. This work fills a gap in the literature and reviews metabolic reactions of hydrolysis and hydarion from the point of views of enzymes, substrates, and reactions.

Viele pharmazeutische Wirkstoffe, Insektizide, Weichmacher für Kunststoffe und andere synthetische Chemikalien enthalten Gruppen wie Ester- oder Amidfunktionen, die in biologischen Medien leicht hydrolysiert werden können. Bis jetzt widmete sich noch kein Buch der Hydratisierung, der Hydrolyse und den dafür verantwortlichen Enzymen, den Hydrolasen, im Zusammenhang mit dem Metabolismus der genannten Verbindungen. Testa und Mayer füllen die Lücke mit diesem Band. Umfassend, aber nicht enzyklopädisch werden im wesentlichen folgende Themen behandelt: biochemische Eigenschaften von Enzymen und Isozymen, Enzym-Substrat-Wechselwirkungen, enzymkatalysierte Reaktionen, Mechanismen und Katalysekreisläufe, Regio- und Stereoselektivität, molekulare Aspekte der Wirkung von Giften und die Inhibition von Enzymen. Der Stoff ist nach den funktionellen Gruppen der abgebauten Verbindungen (Amide, Lactame, Peptide, Carbonsäureester, Ester anorganischer Säuren, Epoxide) geordnet. Sorgfältig ausgew ählte Beispiele veranschaulichen die Argumente.
